Isolation and Characterization of Galactose Epimerase-less Mutant of Salmonella pullorum

Abstract

|
Uridine diphosphate(UDP)-galactose-4-epimerase-less mutant of Salmonella pullorum was isolated after mutagenic treatment with ethidium bromide. When isolated gal E mutant of S. pullorum was grown in the presence of galactose(0.1 W/V), it exhibited marked bacteriolysis in heart infusion broth. The mutant strain was further investigated the changes in the antigenic epitopes of lipopolysaccharides(LPSs) using sodium dodecyl sulfate-polyacrylamide gel electrophoresis and immunoblotting. LPSs from gal E mutant strain had been shown to be composed of only core polysaccharide and shorter O-side chains than those of parent strain of S. pullorum AO150 in SDS-PAGE and in immunoblotting.
